Overview

Epoxide hydrolases (EHs) are enzymes that catalyze the hydrolysis of epoxides into diols. They play a critical role in detoxification, metabolism of endogenous lipid mediators, and regulation of various signaling pathways. Several isoforms exist, including microsomal EH (mEH/EPHX1), soluble EH (sEH/EPHX2), and EPHX3/EHPX4, each with distinct tissue distribution and substrate preferences. sEH, in particular, is a therapeutic target for cardiovascular and inflammatory diseases due to its role in regulating bioactive lipids like EETs. The catalytic mechanism involves a two-step process with nucleophilic attack and water-mediated ester hydrolysis. Dysregulation of EHs is associated with various diseases, making them attractive therapeutic targets.
