Overview

Epstein–Barr virus latent membrane protein 2B (LMP2B) is one of two isoforms (along with LMP2A) encoded by the EBV LMP2 gene. LMP2B is a multi-pass transmembrane protein expressed during EBV latency in infected B cells and in EBV-related malignancies. LMP2B shares most of its sequence with LMP2A but lacks the N-terminal cytoplasmic domain required for LMP2A’s signaling functions. Unlike LMP2A, which actively blocks B-cell receptor (BCR) signaling and promotes B-cell survival, the molecular function of LMP2B remains unclear. Studies show LMP2B may negatively regulate LMP2A by interfering with its aggregation and phosphorylation, potentially modulating the establishment and maintenance of EBV latency. LMP2B does not directly affect BCR signal transduction but can restore normal BCR signaling in cells co-expressing LMP2A, indicating a regulatory interplay between the two isoforms. There is evidence LMP2B is expressed in EBV-associated tumors, suggesting a possible role in viral oncogenesis, but no direct therapeutic targeting or robust biomarker applications have been established for LMP2B.
