Overview

The Epstein-Barr nuclear antigen 1 (EBNA1) peptide–major histocompatibility complex (MHC) is a specialized immunological target presented on the surface of cells infected with the Epstein-Barr virus (EBV). EBNA1 is a DNA-binding protein essential for the persistence and replication of the EBV episome and is uniquely expressed in all EBV-associated malignancies, including nasopharyngeal carcinoma, Burkitt lymphoma, and certain gastric cancers (PMID: 15153531). Although EBNA1 utilizes a glycine-alanine repeat sequence to limit its own proteasomal degradation, defective ribosomal products (DRiPs) allow for the presentation of EBNA1-derived peptides on MHC Class I molecules, such as HLA-A*02:01 (PMID: 10866115). This peptide-MHC complex acts as a molecular signature that allows the immune system, or engineered therapies, to distinguish EBV-positive malignant cells from healthy tissue. Therapeutic interventions targeting this complex include adoptive T-cell therapies like tabelecleucel and experimental TCR-engineered T cells (TCR-T) or TCR-like antibodies (PMID: 31434705). These approaches leverage the high specificity of the TCR-pMHC interaction to induce targeted cell lysis, offering a precision oncology framework for treating EBV-driven diseases while mitigating systemic toxicity.
