Overview

Epstein-Barr nuclear antigen 1 (EBNA1) is a multifunctional viral protein essential for the persistence of the Epstein-Barr virus (EBV) in host cells. It mediates the replication and segregation of the viral episome during mitosis and regulates the transcription of both viral and cellular genes (1.1.1, 1.5.2). EBNA1 is uniquely expressed in all EBV-associated malignancies, including nasopharyngeal carcinoma, Burkitt lymphoma, and Hodgkin lymphoma, making it an ideal target for immunotherapy (1.1.2, 1.4.2). When presented as a peptide on Human Leukocyte Antigen (HLA) molecules, it can be recognized by cytotoxic T lymphocytes (1.1.3, 1.3.4). However, EBNA1 has evolved a glycine-alanine repeat (GAr) domain that inhibits its own proteasomal degradation, thereby reducing its presentation on MHC class I and facilitating immune evasion (1.3.1, 1.3.2). Therapeutic strategies targeting this complex include TCR-engineered T cells and vaccines designed to enhance the immune response against EBNA1-positive cells (1.1.3, 1.4.4).
