Overview

Epstein-Barr nuclear antigen 3A (EBNA-3A) is a viral nuclear protein encoded by the Epstein-Barr virus (EBV), belonging to the EBNA-3 family along with EBNA-3B and EBNA-3C [3][4]. EBNA-3A acts as a transcriptional regulatory protein that interacts with host cellular factors, notably the transcription factor RBP-Jκ (also called CBF-1), a key component of the Notch signaling pathway [1][2]. Through this interaction, EBNA-3A can repress transcription from both viral and host gene promoters, notably downregulating transcription mediated by EBNA-2 and Jκ, and repressing the Cp promoter [1][2]. Functional studies have demonstrated that EBNA-3A is essential for the EBV-mediated immortalization of B lymphocytes in vitro, a process critical to the establishment of latent infection and the development of EBV-driven lymphoproliferative diseases and some cancers, such as certain lymphomas [2][4][5]. EBNA-3A does not have known direct pharmacological inhibitors or approved drugs targeting it, and it is not currently considered a classic therapeutic target such as a receptor, enzyme, or transporter. However, it serves as a potential immunotherapy antigen in experimental settings given its restricted expression in infected and transformed cells.
