Overview

The BMLF1 peptide–MHC complex is a specific molecular assembly presented on the surface of cells infected with the Epstein-Barr virus (EBV) during the lytic phase of the viral life cycle. BMLF1, also known as SM, Mta, or EB2, is an early lytic protein essential for viral mRNA export and gene expression (UniProt P03202). The most well-characterized epitope from this protein is the GLCTLVAML peptide, which is typically presented by the HLA-A*02:01 molecule (PubMed 9151711). This complex serves as a critical target for the host's CD8+ T-cell response, making it a primary focus for the development of adoptive T-cell therapies and TCR-like biologics (PubMed 33116024). In the context of EBV-associated diseases, such as infectious mononucleosis and certain cancers like nasopharyngeal carcinoma, targeting this pMHC complex allows for the selective elimination of virally infected cells. Therapeutic strategies often involve engineered T-cell receptor (TCR) therapies or bispecific antibodies designed to recognize the unique spatial configuration of the peptide within the MHC groove. Because BMLF1 is expressed early in the lytic cycle, it is an ideal target for preventing the spread of the virus and treating EBV-driven malignancies (PubMed 10438931).
