EBI2, also known as GPR183, is a seven-transmembrane G protein-coupled receptor (GPCR) activated by oxysterols like 7α,25-dihydroxycholesterol (7α25HC). It is highly expressed in immune cells, including B cells, T cells, dendritic cells, monocytes/macrophages, NK cells, and granulocytes. EBI2 plays a crucial role in guiding immune cell migration and localization within secondary lymphoid organs, particularly for B cells and dendritic cells. It also modulates cytokine/chemokine production, linking innate and adaptive immunity. Altered EBI2 expression has been linked to autoimmune conditions like SLE and may be involved in CNS immune responses.
Agonism of EBI2 by oxysterols primarily signals through Gi protein to decrease cAMP levels, increase calcium mobilization and activate ERK/MAPK. Activation may also occur through β-arrestin–dependent pathways.
