Epstein-Barr virus (EBV) latent membrane proteins 1 and 2 (LMP1 and LMP2) are critical viral oncogenes that drive the development of EBV-associated malignancies by mimicking host cell signaling receptors (Young & Rickinson, 2004, Nature Reviews Cancer). LMP1 functions as a constitutively active member of the tumor necrosis factor receptor (TNFR) superfamily, specifically mimicking CD40 to activate NF-κB and MAPK pathways, which promote cell survival and proliferation (Dawson et al., 2012, Seminars in Cancer Biology). LMP2, particularly the LMP2A isoform, mimics a constitutively active B-cell receptor (BCR), providing essential survival signals to infected B-cells and preventing them from entering the lytic cycle (Longnecker, 2000, Nature Reviews Immunology). These proteins are expressed in several cancers, including nasopharyngeal carcinoma (NPC) and Hodgkin lymphoma, making them attractive targets for immunotherapy (Lin et al., 2002, Cancer Research). Therapeutic approaches like autologous dendritic cell vaccines pulsed with LMP1/LMP2 peptides aim to stimulate a robust cytotoxic T-lymphocyte (CTL) response to eliminate tumor cells (Chia et al., 2012, Molecular Therapy). By presenting these viral epitopes, the vaccine bypasses the immune evasion strategies typically employed by EBV-positive tumors.
The vaccine utilizes autologous dendritic cells to present specific peptide epitopes of LMP1 and LMP2 to the host immune system, thereby activating and expanding antigen-specific CD8+ and CD4+ T-cells that target and lyse EBV-positive malignant cells (Lin et al., 2002, Cancer Research).
