Overview

Epstein-Barr virus latent membrane protein 1 (LMP1) mRNA is the transcript of the primary oncogene of the Epstein-Barr virus (EBV), a herpesvirus associated with several human malignancies including nasopharyngeal carcinoma and Hodgkin lymphoma (Young & Rickinson, 2004, PMID: 15334130). The mRNA encodes the LMP1 protein, which functions as a constitutively active mimic of the CD40 receptor, triggering signaling pathways such as NF-kappaB, JNK, and PI3K/Akt that promote cell survival, proliferation, and metastasis (Dawson et al., 2012, PMID: 22490334). Because LMP1 is essential for EBV-mediated B-cell transformation and is expressed in many EBV-associated cancers, its mRNA is a high-priority therapeutic target. Strategies to target this mRNA include DNAzymes, such as DZ1, and various siRNA or antisense oligonucleotide formulations designed to silence the gene and induce apoptosis in cancerous cells (Cao et al., 2014, PMID: 24913475). Clinical and preclinical studies focus on using these agents to treat nasopharyngeal carcinoma and EBV-positive lymphomas by reducing the expression of the LMP1 oncoprotein. This targeted degradation not only inhibits tumor growth but also enhances the efficacy of conventional therapies like radiation and chemotherapy (Li & Hong, 2014, PMID: 24511369).
