The Epstein-Barr virus latent membrane protein 1 (LMP1) peptide–MHC complex is a molecular target consisting of LMP1-derived peptides presented by Major Histocompatibility Complex (MHC) molecules, primarily HLA-A*02:01, on the surface of EBV-infected cells (PubMed: 28811304). LMP1 is a principal oncogene of EBV that mimics CD40 signaling to drive B-cell proliferation and survival through the activation of NF-κB and other pathways (UniProt: P03230). As an intracellular protein, LMP1 cannot be targeted by standard antibodies; however, its presentation as a peptide-MHC complex allows for recognition by the cellular immune system and engineered therapies (PubMed: 31434710). This target is highly relevant in EBV-associated malignancies such as nasopharyngeal carcinoma and Hodgkin lymphoma, where LMP1 is consistently expressed (PubMed: 11253396). Therapeutic approaches include TCR-engineered T-cells and TCR-like antibodies, such as ET190L1, which are designed to bind the pMHC complex with high specificity (Eureka Therapeutics). Challenges in targeting this complex include the risk of cross-reactivity with similar human self-peptides and the potential for tumor immune evasion via MHC downregulation (PubMed: 25635007).
