The Epstein-Barr virus (EBV) latent membrane protein 2 (LMP2) peptide-HLA complex is a specialized immunological target found on the surface of EBV-infected malignant cells [PMID: 31534003]. LMP2 is a viral protein expressed during the latent phase of the EBV life cycle, particularly in Latency II and III patterns associated with cancers such as nasopharyngeal carcinoma and Hodgkin lymphoma [PMID: 21148332]. Intracellular processing of LMP2 generates specific peptide epitopes, such as the HLA-A*02:01-restricted CLGGLLTMV, which are presented by Human Leukocyte Antigen (HLA) class I molecules [PMID: 15141015]. These peptide-HLA (pHLA) complexes are recognized by the T-cell receptors (TCRs) of cytotoxic T lymphocytes, marking the cell for destruction [PMID: 33613511]. Because LMP2 is a viral antigen, these complexes are absent from healthy, non-infected human tissues, providing a high degree of tumor specificity [PMID: 25406331]. Therapeutic strategies targeting these complexes include the development of TCR-engineered T-cells (TCR-T) and TCR-like antibodies [ClinicalTrials.gov NCT02739451]. These therapies aim to overcome the immune evasion strategies of EBV-positive tumors by providing a direct mechanism for T-cell recognition and killing [PMID: 30206233].
