Equilibrative nucleobase transporter 1 (SLC43A3) is a member of the solute carrier family 43 and functions as a sodium-independent, purine-selective nucleobase transporter. It mediates the equilibrative transport of extracellular purine bases, such as adenine, guanine, and hypoxanthine, across plasma membranes, particularly in the liver and lung[2][3]. SLC43A3 is also able to transport the purine analog drug 6-mercaptopurine[3]. It does not require sodium or protons for its transport activity and is not involved in nucleoside transport[2]. Beyond nucleobase transport, SLC43A3 may play a regulatory role in fatty acid transport in adipocytes, possibly acting as a positive regulator of fatty acid efflux and a negative regulator of fatty acid uptake[1][3]. SLC43A3 may be relevant in diseases involving purine metabolism and transport, and is also associated with the cellular pharmacokinetics of purine analog drugs like 6-mercaptopurine[3].
