Overview

ER membrane protein complex subunit 10 (EMC10) is a protein-coding component of the endoplasmic reticulum membrane protein complex (EMC), which assists in the biogenesis and membrane integration of multi-pass and tail-anchored proteins within the ER[1][2][3][4]. EMC10 itself is considered an aqueous or luminal subunit of the complex and not a direct membrane insertase. The EMC complex as a whole ensures proper topology and folding of integral membrane proteins, thereby maintaining protein homeostasis and supporting processes like lipid biosynthesis, cellular stress response, and membrane protein quality control[1][2][3][4]. EMC10 has additional reported functions related to promoting angiogenesis and tissue repair following myocardial infarction, via pathways such as p38 MAPK activation[3]. While defects or mutations in EMC10 can contribute to neurodevelopmental disorders and intellectual disability, EMC10 is not an established drug target, and there are no specific therapies or clinically relevant biomarkers associated with it. The EMC family, which includes EMC10, is evolutionarily conserved from yeast to humans and underpins essential aspects of cellular biology linked to ER function and membrane protein management[1][2][3][4].
