Overview

ER membrane protein complex subunit 5 (EMC5, also known as MMGT1 or TMEM32) is an integral component of the eight-subunit endoplasmic reticulum membrane protein complex (EMC), which plays a crucial role in the insertion and correct folding of newly synthesized transmembrane proteins into ER membranes in eukaryotes[3][14][2][6]. The human protein is primarily involved in membrane protein biogenesis by serving as part of a multiprotein insertase that assists the integration of both single-pass and multipass membrane proteins, favoring substrates with weakly hydrophobic or charged transmembrane domains[2][4][14]. Although originally described as a putative magnesium transporter based on sequence similarity, its primary function in humans is as an essential member of the EMC insertase complex, with only indirect evidence supporting a role in magnesium homeostasis[1][3][5][14]. Disruption of EMC5 or the EMC complex in cells leads to impaired membrane protein insertion and may underlie certain genetic diseases connected to protein trafficking defects. No direct interaction with therapeutic drugs or established biomarker status is currently described for EMC5/MMGT1 in clinical settings[3][14].
