ER membrane protein complex subunit 9 (EMC9) is a protein component of the endoplasmic reticulum (ER) membrane protein complex (EMC), which is essential for the biogenesis and insertion of transmembrane domains of multi-pass and tail-anchored membrane proteins into the ER membrane[2][3][8][11]. EMC9 is a cytosolic cap subunit that forms a subcomplex with EMC2 and can be interchangeable with EMC8 in mammals[8][11]. The EMC as a whole acts as a membrane insertase, facilitating the topologically correct, energy-independent integration of nascent membrane proteins into the ER, indirectly impacting diverse cellular processes such as signal transduction and protein trafficking[2][3][11]. Dysfunction of EMT9 or the EMC complex can lead to a variety of developmental and disease states, including neurological defects, visual impairment, and retinoblastoma[2]. EMC9 is not generally considered a direct therapeutic target; rather, it is required for the assembly and proper function of the EMC insertase machinery[2][3][8][11].
