Overview

Erbb2 interacting protein (ERBIN) is a large scaffolding protein of the LAP family containing 17 leucine-rich repeats and a single PDZ domain. ERBIN is primarily known for binding the unphosphorylated form of the ErbB2 receptor (HER2/ERBB2) at Tyr-1248, helping regulate its localization and stabilization in epithelial cells. Through this, ERBIN modulates ERBB2-dependent signaling, potentially acting as a tumor suppressor by disrupting Ras-Raf interaction and negatively regulating oncogenic signaling. ERBIN also inhibits NOD2-mediated NF-κB signaling, thereby moderating inflammation. Clinical relevance is suggested by associations with immunodeficiency and inflammation when ERBIN is deficient, but it is not a current direct drug target.
