Overview

The ERBB3 receptor, also known as HER3, is a member of the EGFR/ERBB family of receptor tyrosine kinases. It binds ligands such as neuregulin-1 (NRG1) and signals primarily through heterodimers, most notably with ErbB2/HER2, activating downstream pathways like PI3K/Akt and MAPK. Although ERBB3 possesses weak intrinsic kinase activity, it plays a crucial role in promoting tumorigenesis and drug resistance in various cancers. Therapeutic strategies targeting ErbB receptors must consider ERBB3's unique biology and its ability to mediate resistance mechanisms.
