Overview

Ergosterol biosynthetic protein 28 homolog (ERG28) is a highly conserved, non-catalytic transmembrane protein involved in sterol biosynthesis pathways across eukaryotes. In yeast, ERG28 serves as a scaffold anchoring the C-4 demethylation enzyme complex (Erg25–Erg26–Erg27) to the endoplasmic reticulum, facilitating efficient ergosterol synthesis[1]. In mammals, ERG28 is homologous to yeast Erg28p and plays an ancillary role in cholesterol biosynthesis; it is co-regulated with core cholesterol enzymes by SREBP-2 and is expressed in most tissues[2]. Knockout studies in human cell lines show that loss of ERG28 significantly decreases cholesterol synthetic flux and cellular cholesterol levels. ERG28 interacts with itself and with key proteins of the demethylation complex (NSDHL, SC4MOL), indicating a structural, platform-like function[2]. Though not a direct enzymatic target, it is implicated in cancer cell proliferation and sterol homeostasis but is not currently a therapeutic target or direct drug interaction point[2].
