Overview

The ERK5 phosphorylation pathway is a mitogen-activated protein kinase (MAPK) signaling cascade, distinct from other MAPKs due to the unique structure and regulatory mechanisms of its central component, ERK5 (also known as BMK1). This pathway plays critical roles in cell proliferation, differentiation, survival, apoptosis, and angiogenesis. The pathway is activated by upstream kinases like MEKK2/MEKK3, which phosphorylate MEK5, which in turn phosphorylates ERK5 at Thr219 and Tyr221 within the TEY motif. Once activated, ERK5 translocates to the nucleus and phosphorylates transcription factors such as SAP1, c-FOS, and MEF2, regulating gene expression. Dysregulation of the ERK5 pathway contributes to tumorigenesis and resistance to conventional therapies, making it an emerging anti-cancer target.
