Erythrocyte membrane protein band 4.1-like 3 (EPB41L3)

Target
EPB41L3
Molecular classification
Cytoskeletal adapter protein, Structural constituent of the cytoskeleton, FERM domain-containing protein (FERM = *Four-point-one, Ezrin, Radixin, Moesin*; involved in linking membrane proteins to the cytoskeleton)[3]
01

Overview

Erythrocyte membrane protein band 4.1-like 3 is a cytoskeletal adapter protein expressed in multiple tissues, including the brain, lung, kidney, and intestine[3][4]. It contains a FERM domain, allowing it to link the plasma membrane to the cytoskeleton and interface with transmembrane proteins such as CD44[3]. EPB41L3 regulates cell adhesion, inhibits cell proliferation, and promotes apoptosis by modulating caspase and protein arginine methyltransferase activity[1][2][4]. Knockout studies indicate roles in nervous system development and myelin organization[3]. EPB41L3 functions as a tumor suppressor, whose reduced expression (often through promoter hypermethylation) contributes to cancer development and poor prognosis[2][4]. Molecular interactions, epigenetic regulation, and its methylation status are under investigation for cancer diagnosis and therapeutic intervention. Note: There are no widely approved drugs targeting EPB41L3 directly. Research is ongoing into small molecules that may modulate EPB41L3 function or its interactions, with a focus on cancer[3]. Promoter methylation status and protein expression are in use as clinical biomarkers for cancer prognosis[2][4].

Mechanism of action

Potential future drugs may act by restoring or enhancing EPB41L3 tumor suppressor activity, modulating its methylation status, or altering its interaction with CD44 or cytoskeletal partners[2][3]

03

Biological functions

Maintenance of cell-cell contact and adhesionLinking the plasma membrane to the cytoskeletonRegulation of cell proliferationPromotion of apoptosisModulation of cell cycle (G2/M arrest via CDK1 pathway[2])Regulation of protein arginine methyltransferases (PRMT3, PRMT5)[1][3][4]Maintenance of paranodal junctions in neuronsProtein localization to axon[4]
04

Disease associations

Cancer (including non-small cell lung cancer, esophageal squamous cell carcinoma, meningioma, thyroid gland adenocarcinoma)[1][2][4]Neurodegenerative disorders (role in nervous system development, Alzheimer’s disease correlation, Charcot-Marie-Tooth disease, leukoencephalopathy)[3][1]Sensorimotor and neurogenetic syndromes (spastic paraplegia, ataxias)[1]Other (Pelizaeus-Merzbacher disease[1])
05

Safety considerations

None documented for direct targeting; general challenges include tissue-specific expression, redundancy with paralogs like EPB41L1, and incomplete functional characterization[3][4]
06

Biomarkers

Promoter methylation of EPB41L3 is a biomarker for poor prognosis in various cancers, including non-small cell lung carcinoma and esophageal squamous cell carcinoma[2]Protein expression levels as biomarker in meningioma[4]
