Overview

Escherichia coli F4 fimbriae, historically known as K88, are filamentous surface appendages found on enterotoxigenic Escherichia coli (ETEC) strains that are a primary cause of diarrhea in neonatal and weaned piglets [Merck Veterinary Manual]. These fimbriae are complex protein structures composed of multiple subunits, with the FaeG protein serving as the major subunit and primary adhesin responsible for recognizing specific carbohydrate moieties on porcine intestinal receptors [UniProt: P02971]. By facilitating stable attachment to the small intestinal epithelium, F4 fimbriae enable ETEC to colonize the gut and deliver enterotoxins that disrupt electrolyte balance and cause fluid loss [PMID: 15505000]. Given their essential role in the early stages of infection, F4 fimbriae are key targets for veterinary vaccines, including live-attenuated and subunit formulations, as well as novel anti-adhesive strategies [PMID: 23433371]. Therapeutic interventions typically aim to elicit a robust mucosal immune response to block bacterial attachment or physically interfere with the interaction between the fimbriae and host cell receptors to prevent disease progression [PMID: 19146601].
