The Escherichia coli K99 fimbrial antigen, also known as F5, is a major virulence factor of enterotoxigenic Escherichia coli (ETEC) that causes severe, often fatal, diarrhea in neonatal ruminants and swine (Moon et al., 1977). It is a proteinaceous, filamentous surface appendage composed primarily of the FanC subunit, which facilitates the adherence of the bacteria to the intestinal epithelium by binding to N-glycolylneuraminic acid-containing receptors (Kyogashima et al., 1989; UniProt P02971). This attachment is a mandatory prerequisite for the bacteria to colonize the small intestine and deliver heat-stable enterotoxins (STa), which disrupt electrolyte balance and lead to significant economic losses in the livestock industry (Acosta-Dibarrat et al., 2007). Therapeutic and preventive strategies focus on blocking this adhesion through the use of maternal vaccines or the oral administration of monoclonal antibodies to neonates (Castrucci et al., 1988). Because the K99 antigen is highly conserved among pathogenic ETEC strains, it serves as an effective target for broad-spectrum veterinary immunotherapies and diagnostic assays (Mainil, 2013). Current commercial products like ScourGuard and Guardian utilize this antigen to induce protective colostral immunity in pregnant dams (Zoetis; Merck Animal Health).
Prevention of bacterial attachment to the intestinal mucosa by inducing or providing specific antibodies that bind to the fimbrial subunits, thereby blocking colonization and subsequent toxin delivery.
