Escherichia coli O-antigen polysaccharides are complex carbohydrate structures that form the outermost part of the lipopolysaccharide (LPS) layer on the surface of Gram-negative E. coli bacteria. The O-antigen is a key determinant of serotype and plays a crucial role in immune evasion, virulence, and host-pathogen interactions. They typically comprise 10–25 repeating units, each containing two to seven sugar residues. Over 180 different E. coli O-serotypes have been described, highlighting the structural diversity.
