The Escherichia coli O25B O-antigen epitope is a specific carbohydrate structure found within the lipopolysaccharide (LPS) layer of certain extraintestinal pathogenic E. coli (ExPEC) strains, most notably the multi-drug resistant ST131 clone. This O-antigen serves as a critical virulence factor by protecting the bacterium from host complement-mediated killing and facilitating colonization of host tissues. Due to its high prevalence in antibiotic-resistant infections, particularly urinary tract infections and neonatal sepsis, it has become a primary target for novel therapeutic interventions. Current drug development efforts focus on using this epitope as a vaccine candidate or as a target for monoclonal antibodies to provide passive immunity. By targeting the O25B epitope, these therapies aim to enhance opsonophagocytosis and neutralize the pathogen's ability to cause systemic infection.
