Overview

The Escherichia coli outer membrane is a complex, asymmetric lipid bilayer that serves as the primary protective barrier for the bacterium (Wikipedia, 2024). It is composed of an inner leaflet of phospholipids and an outer leaflet dominated by lipopolysaccharides (LPS), which are essential for maintaining structural integrity and resisting environmental stressors, including many antibiotics (NIH, 2023). This surface also contains various outer membrane proteins (OMPs) such as porins, which facilitate nutrient transport, and virulence factors like pili that mediate adhesion to host tissues (Mirage News, 2024). As a therapeutic target, the outer membrane is the site of action for polymyxins, which bind to LPS and disrupt membrane stability, leading to cell death (NIH, 2018). Emerging therapies also target specific assembly machineries within the membrane, such as the Bam and Lpt complexes, to compromise the bacterial envelope (NIH, 2023). However, targeting the outer surface can trigger the massive release of endotoxins (LPS), potentially leading to severe systemic inflammation and septic shock in patients (NIH, 2023).
