Overview

ESF1 nucleolar pre-rRNA processing protein homolog (ESF1) is an RNA-binding protein encoded by the *ESF1* gene (HGNC:15898), located on chromosome 20. ESF1 plays a role in rRNA processing and is implicated in the regulation of basal transcription, possibly constituting a novel regulatory system. It localizes predominantly to the nucleolus and is associated with RNA metabolic pathways. ESF1 is upregulated in estrogen receptor-positive breast cancer, where its expression correlates with tumor proliferation and poor patient prognosis. Knockdown of ESF1 inhibits colony formation, increases apoptosis, and impairs wound healing in breast cancer cell models. Diseases linked to ESF1 include familial hypocalciuric hypercalcemia type I. At present, ESF1 is not associated with any known drug interactions or targeted therapies, but its association with cancer proliferation suggests potential as a future therapeutic target.
