Overview

Essential outer membrane proteins (OMPs) are critical components of the Gram-negative bacterial cell envelope, responsible for maintaining structural integrity and mediating the transport of nutrients, waste, and signaling molecules. Key members of this group include the Beta-barrel Assembly Machinery (BAM) complex, specifically BamA, and the Lipopolysaccharide Transport (LPT) machinery, specifically LptD. These proteins are indispensable for bacterial survival because they facilitate the correct folding and insertion of other OMPs and the translocation of lipopolysaccharides to the cell surface. Due to their exposure on the bacterial surface and lack of human homologs, they are highly attractive targets for novel antibiotics. Drugs targeting these proteins, such as Zosurabalpin and Darobactin, work by disrupting the assembly of the outer membrane, which leads to increased membrane permeability and rapid cell lysis. This mechanism is particularly effective against multi-drug resistant (MDR) pathogens like Acinetobacter baumannii and Pseudomonas aeruginosa.
