ETS variant transcription factor 3 (ETV3) is a member of the ETS family of transcription factors, characterized by a conserved ETS DNA-binding domain that recognizes core GGA(A/T) motifs in target DNA[1][4]. ETV3 acts primarily as a transcriptional repressor, modulating gene expression programs involved in cell growth, differentiation, and immune function. Specifically, ETV3 contributes to growth arrest during terminal macrophage differentiation by repressing genes downstream of Ras signaling and is also involved in the regulation of monocyte differentiation, particularly influencing the balance between macrophage and dendritic cell lineage outcomes[2][4]. Its action includes the repression of interferon-stimulated genes and direct inhibition of macrophage differentiation through suppression of MAFB expression. While ETV3 and its closely related factor ETV6 are molecular regulators of immune cell differentiation and have been proposed as potential modulators of inflammatory responses, there is currently no direct evidence supporting the use of drugs targeting ETV3 in clinical practice[2][4].
