ETS variant transcription factor 4 (ETV4) is a member of the ETS family of transcription factors, particularly the PEA3 subfamily. It functions primarily as a DNA-binding transcriptional activator, playing crucial roles in embryonic development, keratinocyte differentiation, and regulation of cellular proliferation. ETV4 acts as a molecular transducer that links mechanical microenvironments (such as cell crowding) to gene expression, impacting lineage specification in human embryonic stem cells. Pathologically, ETV4 overexpression is associated with several cancers (notably Ewing sarcoma), and it is implicated in cancer progression and resistance to certain therapies via maintenance of MYC expression and modulation of the MAPK signaling pathway. ETV4's expression and function in cancer and stem cell biology make it an important molecular marker and a potential but challenging therapeutic target.
