Overview

Eukaryotic translation elongation factor 1 delta (EEF1D) is a key subunit of the elongation factor-1 complex that catalyzes guanine nucleotide exchange for eEF1A, facilitating the delivery of aminoacyl-tRNAs to the ribosome during protein synthesis. It exists as multiple isoforms: the short isoforms (canonical translation factors) are broadly expressed and crucial for protein biosynthesis, cell cycle progression, and proliferation, while the long isoform (eEF1BδL) acts as a heat-shock responsive transcription factor, mainly in brain and testis, implicated in stress response and neurodevelopment. EEF1D's dysregulation is correlated with tumorigenesis, poor prognosis in cancer, cell proliferation control, and regulation of the Akt-mTOR/PI3K pathways as well as epithelial-mesenchymal transition (EMT). Beyond translation, EEF1D interacts with viral proteins, modulating host protein synthesis, and has roles in nervous system development and disease.

Mechanism of action

For hypothetical drugs: Inhibition or downregulation of EEF1D may suppress cell proliferation, tumor growth, and cell cycle progression. Modulation of its interaction with viral proteins could inhibit viral replication. Targeting post-translational modifications (e.g., phosphorylation on Ser133) regulates cell cycle and proliferation.
