Overview

Eukaryotic translation elongation factor 1 gamma (EEF1G) is a protein encoded by the EEF1G gene in humans and functions as a subunit of the elongation factor-1 (eEF1) complex, which is central to the elongation phase of protein synthesis[3][5]. The elongation factor-1 complex delivers aminoacyl tRNAs to the ribosome during translation[1][2][5]. EEF1G contains an N-terminal glutathione S-transferase (GST) domain, thought to be involved in assembling multisubunit complexes that include other elongation factor subunits and aminoacyl-tRNA synthetases, and may also anchor the eEF1 complex to cellular structures[2][4]. As part of the eEF1B complex (with eEF1B2 and eEF1D), EEF1G assists in reactivating eEF1A by facilitating GDP to GTP exchange, indirectly supporting protein synthesis fidelity and efficiency[1][4]. Functionally, EEF1G has been implicated in viral protein translation, viral replication (notably influenza and vesicular stomatitis virus), and has roles in anchoring or scaffolding protein complexes beyond canonical translation functions[1]. Variations in EEF1G expression or structure can affect cell proliferation and stability of the eEF1B complex[1]. Its expression and mutational status have been connected to cancer biology and responses to viral infections, but it is not currently a direct drug target, biomarker, or associated with specific therapeutic safety concerns[4].
