EIF2AK1 is a protein kinase that regulates protein synthesis in response to cellular stress by phosphorylating eIF2α, inhibiting translation initiation. It is activated by oxidative stress, heme deficiency, osmotic shock, and heat shock. Dysregulation is linked to leukoencephalopathy and hypomyelinating leukodystrophy.
Phosphorylation of eIF2α/EIF2S1 at Ser48 and Ser51, preventing recycling of eIF2 and inhibiting ternary complex formation.
