Eukaryotic translation initiation factor 2B (eIF2B) is a crucial multisubunit protein complex that acts as the guanine nucleotide exchange factor (GEF) for eukaryotic initiation factor 2 (eIF2). It catalyzes the exchange of GDP for GTP on eIF2, regenerating active eIF2-GTP, which is essential for protein synthesis initiation. eIF2B activity is regulated by phosphorylation and other cellular signals, and mutations in eIF2B subunits can cause leukoencephalopathy with vanishing white matter (VWM). It represents a potential therapeutic target in neurodegenerative diseases.
