Overview

Eukaryotic translation initiation factor 2D is a cytoplasmic translation factor that facilitates the recruitment and delivery of both initiator and noninitiator aminoacyl-tRNAs to the P-site of the eukaryotic ribosome in a GTP-independent manner[3][2][4]. Unlike canonical eIF2, eIF2D can deliver non-Met (elongator) tRNAs into the P-site and promote the release of deacylated tRNA and mRNA from recycled 40S subunits following ABCE1-mediated dissociation of ribosomal complexes[4]. EIF2D contains PUA and SUI1 domains, with roles in RNA binding and potential ribosome conformation regulation[2]. It has been previously referred to as ligatin (LGTN), and is sometimes called hepatocellular carcinoma-associated antigen 56 (HCA56), though this designation is historical[3][2]. Several transcript variants encode different isoforms[3]. While EIF2D is not strictly essential for general translation, it may play roles under specific or stress-related cellular conditions, or with specific RNAs[2]. Disease associations include links to autism spectrum disorder and certain neurodegenerative conditions, though these are not universally accepted therapeutic targets[3]. No interacting drugs, related mechanisms of action, or published safety concerns are documented for this protein.
