Overview

Eukaryotic translation initiation factor 3 subunit C-like (EIF3CL) is a core protein component of the eukaryotic translation initiation factor 3 (eIF3) complex, a large multiprotein assembly essential for the initiation of protein synthesis in eukaryotic cells. EIF3CL is nearly identical to EIF3C, a closely related paralog. The eIF3 complex binds the 40S ribosomal subunit and acts as a scaffold to recruit other initiation factors and the initiator tRNA, facilitating assembly of the 43S pre-initiation complex. This complex promotes mRNA recruitment and scanning for the AUG start codon, and prevents premature joining of the 60S ribosomal subunit. Additionally, eIF3 components help recycle ribosomal complexes after translation termination and regulate specific subsets of mRNA translation. While dysregulation of eIF3 subunits has been implicated broadly in cancer, EIF3CL itself is primarily recognized for its conserved, essential role in general protein synthesis and has been associated with autism spectrum disorder[1][2][5][6].
