Eukaryotic translation initiation factor 3 subunit E (EIF3E) is a protein that forms part of the eukaryotic translation initiation factor 3 (eIF3) complex, which is essential for the initiation of protein synthesis in eukaryotic cells[1][3][7]. EIF3E contributes to multiple molecular processes including RNA binding, positive regulation of translation, and formation of the 43S preinitiation complex. As a member of the eIF3 complex, it associates with the 40S ribosomal subunit, stimulates mRNA recruitment, and is involved in the scanning and selection of AUG start codons for translation initiation. Moreover, EIF3E is implicated in nonsense-mediated mRNA decay and may regulate cell proliferation, cell cycle progression, differentiation, and apoptosis by targeting select mRNAs[1][7]. Dysregulation or mutation of EIF3E has been linked to several cancers, including breast, lung, colorectal, prostate cancer, and melanoma[2]. At present, specific drugs targeting EIF3E are not established, and its role as a translation factor makes it a challenging therapeutic target due to the potential for broad effects on protein synthesis.
