Overview

Eukaryotic translation initiation factor 3 subunit J (EIF3J) is a core component of the eukaryotic translation initiation factor 3 (eIF3) complex, which orchestrates the initiation of protein synthesis in eukaryotic cells[2][4]. EIF3J binds the eIF-3 complex through EIF3B and locates at the aminoacyl site and mRNA entry channel of the 40S ribosomal subunit, placing it in the ribosomal decoding center[1][3]. It modulates access to the mRNA-binding cleft, interacts with other initiation factors (notably eIF1A), and is essential for multiple steps of initiation, including the recruitment of mRNA to the ribosome, assembly of the pre-initiation complex, and regulation of scanning for the start codon[1][2][3]. EIF3J also cooperates in translation termination by stimulating peptidyl-tRNA hydrolysis in conjunction with release factors and participates in ribosomal recycling[3]. While critical for correct translational control, it is not a direct therapeutic target nor a clinical biomarker, but its indirect influence on cell proliferation, differentiation, and apoptosis links it to fundamental cellular processes[2][3].
