Eukaryotic translation initiation factor 4 gamma 3 (EIF4G3) is a protein that serves as a core component of the eIF4F complex, a key regulator of cap-dependent translation initiation in eukaryotic cells[2][7]. EIF4G3 acts as a large scaffold protein that mediates the assembly of multiple translation initiation factors, including eIF4E (cap-binding protein), eIF4A (RNA helicase), and eIF3 (ribosome recruitment factor), thereby facilitating the recruitment of mRNA to the ribosome, unwinding of the 5' UTR mRNA secondary structure, and overall initiation of protein synthesis. EIF4G3 is an RNA binding protein, has important roles in the regulation of cell proliferation, differentiation, and is expressed in multiple tissues with prominence in the central nervous system and the testes[4][5][7]. Its activity is tightly regulated, including by microRNAs such as miR-520c-3p, which suppress EIF4G3 translation and thereby reduce protein synthesis and cell proliferation, underlying its potential relevance in cancer biology[2]. Dysfunction or altered expression of EIF4G3 has been associated with several diseases, especially certain cancers such as gastric cancer[2]. There are currently no known drugs or pharmacological inhibitors that specifically target EIF4G3.
