Overview

Eukaryotic translation initiation factor 4A (eIF4A) is a highly conserved, essential protein involved in the initiation phase of eukaryotic mRNA translation. It functions as an ATP-dependent RNA helicase and is a core component of the eIF4F complex, which mediates the recruitment of ribosomes to mRNA by unwinding secondary structures in the 5' untranslated region (UTR) of mRNAs. It has three isoforms: EIF4A1, EIF4A2, and EIF4A3. EIF4A1 and EIF4A2 are functionally interchangeable and essential for cap-dependent translation. EIF4A3 does not substitute for the other isoforms, and may inhibit translation under certain conditions. Its dysregulation has been linked to cancer progression.
