Overview

Eukaryotic translation initiation factor 4E-binding protein 1 is a small regulatory protein that inhibits cap-dependent mRNA translation by binding to eukaryotic translation initiation factor 4E (eIF4E)[1][6][7]. Under physiological conditions, 4E-BP1 represses translation by preventing assembly of the eIF4F complex, which is required for recruiting ribosomes to messenger RNAs. Upon phosphorylation by kinases downstream of mTOR and other signaling pathways, 4E-BP1 dissociates from eIF4E, relieving translational repression and promoting protein synthesis. Dysregulation or hyperphosphorylation of 4E-BP1 is frequently observed in cancers and is linked to poor outcome due to enhanced translation of growth-promoting and oncogenic mRNAs[1][2]. 4E-BP1 and its regulators have emerged as promising therapeutic targets for controlling aberrant protein synthesis in oncology and metabolic diseases.
