Overview

Eukaryotic translation initiation factor 4E nuclear import factor 1 (EIF4ENIF1), also known as 4E-T, is a nucleocytoplasmic shuttle protein that regulates the localization and activity of the translation initiation factor eIF4E. It binds eIF4E, mediating its nuclear import via the importin alpha-beta complex—a process essential for proper control of mRNA translation. EIF4ENIF1 resides predominantly in the cytoplasm but can shuttle into the nucleus. In the cytoplasm, it plays a crucial role in the formation and function of processing bodies (P-bodies), coordinating the storage, translational repression, and degradation of mRNAs. By interacting with RNA-binding proteins and protein complexes such as CCR4-NOT, EIF4ENIF1 promotes deadenylation (tail shortening) and prevents decapping of mRNAs, protecting them from degradation and maintaining translationally inactive pools. It is critically involved in oocyte development and female fertility, with loss-of-function mutations associated with primary ovarian insufficiency. Mutations in EIF4ENIF1 are also found in some autism spectrum disorder cases. EIF4ENIF1 does not currently have known therapeutically targeted drugs or biomarker applications, and there are no prominent safety concerns described in the literature.
