Even-skipped homeobox 1 (EVX1) is a transcription factor belonging to the homeobox gene family, containing a DNA-binding homeodomain highly homologous to the Drosophila even-skipped (eve) segmentation gene. EVX1 functions primarily as a transcriptional repressor during embryogenesis, playing key roles in the specification of neuronal cell types and normal blastocyst development. In prostate cancer, EVX1 is frequently hypermethylated, which is associated with reduced gene expression and poor clinical outcomes, making its methylation a potential biomarker for disease progression. Mutations in EVX1 are linked to congenital limb and urogenital malformations. EVX1 does not represent a common therapeutic target such as a receptor or enzyme, but has mechanistic and biomarker importance in development and cancer.
