EWS-FLI1 is an oncogenic fusion protein pathognomonic for Ewing sarcoma, resulting from a chromosomal translocation, most commonly t(11;22)(q24;q12), which fuses the N-terminal domain of the EWSR1 gene to the C-terminal DNA-binding domain of the FLI1 gene. It functions as an aberrant transcription factor, dysregulating genes involved in cell proliferation, differentiation, and survival. The fusion protein is localized to the nucleus and represents a therapeutic target for Ewing sarcoma.
