Ewing tumor-associated antigen 1 (ETAA1) is a critical protein involved in the cellular response to DNA replication stress and the maintenance of genomic integrity. It serves as a direct activator of the ATR (ataxia telangiectasia and Rad3-related) kinase, which is a master regulator of the DNA damage checkpoint. ETAA1 is recruited to stalled replication forks through its interaction with the replication protein A (RPA) complex, where it stimulates ATR activity independently of the TOPBP1 pathway. Originally identified as a tumor-specific antigen in the Ewing family of tumors, ETAA1 has since been recognized for its broader role in various malignancies, including pancreatic and liver cancers. In the context of oncology, ETAA1 is a key component of the replication stress response pathway, making it a significant factor in the efficacy of ATR inhibitors and other DNA damage response (DDR) targeted therapies currently under development.
