Exocyst complex component 3 is a cytosolic protein encoded by the EXOC3 gene in humans. It is a core member of the exocyst complex, which consists of eight unique subunits (EXOC1–8) that work together to tether secretory vesicles for targeted fusion at specific sites of the plasma membrane. This process is essential for directed exocytosis, maintenance of epithelial cell surface polarity, and regulation of cell trafficking pathways. In platelets, EXOC3 controls granule secretion and the trafficking of glycoprotein receptors, influencing thrombosis and hemostatic function, but does not exhibit redundancy with its paralogs in this cell type[1][2][3][5][6]. Deficiency in EXOC3 impairs vesicle trafficking dynamics, leading to functional defects in cellular secretion and signaling pathways.
