Exogenous ligands are chemical substances that originate outside of an organism's body and are capable of binding to and modulating the activity of specific biological targets, such as receptors, enzymes, or transporters (StatPearls, 2023). This broad category includes pharmaceutical drugs, environmental toxins, dietary components, and pathogen-derived molecules like lipopolysaccharides. Unlike endogenous ligands, which are produced internally (e.g., hormones or neurotransmitters), exogenous ligands are introduced into the system via ingestion, inhalation, or other routes of exposure (PubMed, 2021). In pharmacology, these molecules are designed or identified based on their ability to mimic or inhibit the effects of natural ligands to achieve a therapeutic outcome. Because the term 'Exogenous ligands' describes the agents that act upon a target rather than a specific protein, gene, or biological structure itself, it is not considered a valid therapeutic target in a clinical or molecular sense. Instead, it serves as a functional classification for xenobiotics and therapeutic agents that interact with the body's biochemical pathways (NIH, 2022).
Exogenous ligands act as external agents that bind to endogenous receptors, enzymes, or transporters to modulate biological signaling pathways (StatPearls, 2023).
