Overview

Exosome component 10 (EXOSC10) is a catalytically active 3'-5' exoribonuclease and a core subunit of the multi-protein RNA exosome complex, primarily located in the nucleus. EXOSC10 plays a crucial role in the 3'-5' processing and degradation of many RNA species, including rRNA, small nucleolar RNA (snoRNA), and defective mRNA[1][5]. It is involved in various cellular processes, such as ensuring the quality of RNA, regulating the maturation and elimination of RNA intermediates, controlling mRNA turnover, and participating in DNA repair via homologous recombination[1]. EXOSC10 is also implicated in cell proliferation, embryonic development, oogenesis, and spermatogenesis[1]. Autoantibodies directed against EXOSC10 (also known as PM/Scl-100) serve as important biomarkers in specific autoimmune disorders, including scleroderma and polymyositis[1]. No drugs currently target EXOSC10 directly. Although part of a major RNA degradation machinery associated with several cellular functions, EXOSC10 itself is not considered a canonical therapeutic target, and loss of function is likely incompatible with cell survival[1][5].
