Overview

Plasmodium export protein 1 (EXP1) is a critical, highly conserved membrane protein located in the parasitophorous vacuole membrane (PVM) of Plasmodium falciparum and other malaria-causing parasites [1, 2]. It serves a dual function essential for parasite survival: it acts as a membrane-bound glutathione S-transferase (mGST) that detoxifies host-derived heme and is vital for maintaining the structural integrity and protein organization of the PVM [2, 3]. Studies have shown that EXP1 is necessary for the proper localization of the PTEX translocon and for the uptake of nutrients from the host cell [3, 4]. Because EXP1 is essential for the parasite's asexual blood stage and lacks significant homology with human proteins, it represents a potent target for antimalarial drug development [2]. Additionally, EXP1 has been implicated in the parasite's response to artemisinin, with altered expression levels correlating with drug sensitivity [2].
