Extended synaptotagmin-2 (ESYT2) is a multi-domain endoplasmic reticulum (ER) membrane protein that acts as a tether between the ER and plasma membrane (PM)[2][4][5][6]. It contains a synaptotagmin-like mitochondrial-lipid-binding protein (SMP) domain and multiple C2 domains, which enable regulated contact and lipid transfer between ER and PM[1][2][3][4][5]. ESYT2, together with E-Syt1 and E-Syt3, forms the extended synaptotagmin protein family found in animals and is orthologous to tricalbins in yeast[2][4][5]. Its functions critically depend on calcium binding and the specificity of C2 domains, including Ca2+-dependent and -independent interactions with phospholipids[2][3][5]. ESYT2 is essential for maintaining ER–PM contact site architecture and involved in the transfer of glycerophospholipids, but is not currently established as a direct therapeutic target or implicated in major disease processes[2][5].
