Overview

Extended synaptotagmin-3 (ESYT3) is a membrane-anchored protein of the endoplasmic reticulum (ER) that tethers the ER to the plasma membrane (PM) via its C2 domains and mediates lipid transfer between these membranes through its synaptotagmin-like mitochondrial-lipid binding protein (SMP) domain[1][2][3]. ESYT3 forms part of the extended synaptotagmin family, which participates in constitutive and regulated ER–PM contact formation, helping maintain plasma membrane lipid homeostasis and facilitating inter-organelle communication in eukaryotic cells[1][2]. ESYT3 is predicted to bind calcium ions and phospholipids, and plays a non-redundant role in specific cellular contexts for organizing membrane contact sites; however, knockout studies in mammals have so far shown minor or context-dependent phenotypes[4][5]. There is currently no evidence of ESYT3 serving as a direct therapeutic target, nor is there documentation of marketed drugs acting on it.
